The homoheptameric PANX1 (Pannexin-1) channel (Ruan et al. 2020, Zhang et al. 2021, Kuzuya et al. 2022) transports ATP from the cytosol to the extracellular region in response to mechanical stimuli such as membrane stretching (Bao et al. 2004, Wang et al. 2016). In osteocytes, PANX1 channels are activated by elevated cytosolic calcium (Locovei et al. 2006) and directly interact with the ATP-gated cation channel P2RX7, which imports calcium (Locovei et al. 2006), therefore P2RX7 and PANX1 may constitute a positive feedback loop.
