The purinergic receptor P2RY2 (P2Y2, P2U) binds ATP (Erb et al. 1995, Nicholas et al. 1996) and activates signaling by the G protein alpha subunit Gq or G11 (Gq/11) (inferred from rabbit homologs in Murthy and Makhlouf 1998). Both P2RY2 and its associated G protein alpha subunit, Gq/G11, are required for elevated cytosolic Ca2+ and activation of nitric oxide synthase 3 (NOS3, eNOS) in response to shear stress (Wang et al. 2015, 2016). P2RY2 and Gq/11 may act through phospholipase C beta (PLCbeta) to activate NOS3.
