Several cellular RNA species act as PKR inhibitors, supporting general protein translation. For instance, VTRNA2-1 (ns886, miR-886), a noncoding vault RNA that is transcribed by RNA polymerase III,, takes part in host cell regulation of PKR activity. Its two conformers bind to PKR and can either inhibit or weakly enhance PKR dimerization and activation (Lee et al, 2011; Jeon et al, 2012; Calderon & Conn, 2015). VTRNA2-1 activity has been implicated in pathological progress of oncogenesis (Lei J et al. 2017; Gao W et al. 2022) while several studies identified a tumor suppressive role of VTRNA2-1 (Fort RS et al. 2018, 2019; Yang R et al. 2021).
