KDM6B demethylates histone H3 trimethyllysine-27 (H3K27me3, lysine-28 of the preprotein) to yield histone H3 monomethyllysine-27 (H3K27me1) (Agger et al. 2007, Hong et al. 2007, Lan et al. 2007, and inferred from the mouse homolog). KDM6B is associated with loss of H3K27me3 in cleavage stage mouse embryos (Yang et al. 2016) and is required for loss of H3K27me3 in cleavage stage bovine embryos (Canovas et al. 2012).
