Human respiratory syncytial virus (RSV) nonstructural protein (NS2 or 1B) interacts with the N-terminal caspase activation recruitment domain (CARD) of DDX58 (RIG-I) upon co-expression in human embryonic kidney 293T (HEK293T) cells (Ling Z et al. 2009). Viral 1B also co-precipitates with endogenous DDX58 during RSV infection in human lung carcinoma (A549) cells. The binding of viral 1B (NS2) is believed to prevent CARD-CARD interactions between DDX58 and MAVS, thereby inhibiting DDX58 (RIG-I)-mediated induction of IFN. In summary, hRSV 1B acts as a multifunctional IFN antagonist, helping the virus to evade host innate immunity (Ling Z et al. 2009; Pei et al, 2021).
