COT (MAP3K8) phosphorylates NIK (NF-κB-inducing kinase), a key step in activating the non-canonical NF-κB signaling pathway. This phosphorylation enhances NIK's role in activating the IKKα complex, leading to the processing of NF-κB2 (p100) into its active form, p52 (Wittwer and Schmitz 2008).
