Priming phosphorylation of the transmembrane protein Smoothened (SMO) leads to additional phosphorylation by Casein kinase I alpha (CKIalpha) at Ser670, Ser673, Ser690, Ser693, Ser743, and Ser746. Even though SMO has been shown to be explicitly phosphorylated by CKIalpha, phosphorylation of these residues by other CK1 isoforms cannot be ruled out. In addition, other residues are probably phosphorylated by unidentified kinases.
