Shh is released from the plasma membrane by ADAM17-mediated cleavage
Coexpression of mouse Shh and human ADAM17 in Bosc23 cells results in the release of a soluble Shh Np lacking the C-terminal cholesterol moiety and the N-terminal palmitoylation. Hh shedding is dependent on ADAM17, as inactivation of the metalloprotease by mutation of the catalytic site or by siRNA-mediated depletion abrogates the ligand release (Dierker et al, 2009).
