Tcf7l2:Ctnnb1, Foxa2, Mixl1, and Gata4 bind the Sox17 gene
Sox17 is a transcription factor whose expression in mice is activated in prospective definitive endoderm cells before intercalation of the definitive endoderm cells with the visceral endoderm (Viotti et al. 2014). Sox17 is necessary for gut endoderm formation in mice (Viotti et al. 2014). Lef1:Cttnb1 from Wnt signaling binds and activates the Sox17 promoter (Engert et al. 2013). Tbxt, whose expression initiates in the primitive streak, also binds the Sox17 promoter (Lolas et al. 2014). Mixl1 binds the Sox17 promoter and activates expression (Lim et al. 2009). Foxa2 and Gata4, endodermal transcription factors, bind a distal enhancer element that may interact with a downstream transcription start site of the Sox17 gene (Trinh et al. 2022).
