Expression of Noto gene in axial mesoderm
Expression of homeobox protein Noto is activated by Foxa2 and Tbxt (T, Brachyury) (Abdelkhalek et al. 2004, Alten et al. 2012), however, when Foxa2 and Foxa1 were knocked out later in development at stage E7.5, expression of Noto was relatively normal until stage E10.5 when expression of Noto was lost (Maier et al. 2013). Tead2:Yap and Lef1:Ctnnb1 or Tcf7:Ctnnb1 activate the Noto promoter in vitro but appear to be dispensable for activation in the node and notochord in vivo (Alten et al 2012). Noto represses paraxial mesoderm fate and maintains axial mesoderm fate (Yamanaka et al. 2007). Knockout of Noto in mice demonstrates that Noto regulates node morphogenesis, notochordal ciliagenesis, and left-right patterning (Beckers et al. 2007). Part of this regulation is accomplished by the activation of cilia-related genes in the node and posterior region of the notochord by Noto (Beckers et al. 2007).
