Expression of Foxa2 in the anterior primitive streak
Foxa2 is first expressed at the anterior end of the primitive streak and then persists in notochord and definitive endoderm (Ang et al. 1993, Sasaki and Hogan 1993, Monaghan et al. 1993, Ruiz et al. 1993, Besnard et al. 2004). The Foxa2 gene contains enhancers that activate and enhance expression in the node and notochord (Sasaki and Hogan 1996, Nishizaki et al. 2001). Nodal signaling activates Smad2, Smad3 which then interact with Foxh1 at the Foxa2 promoter to activate transcription of Foxa2 (Yamamoto et al. 2001, Hoodless et al. 2001). Tead proteins also bind the Foxa2 promoter and interact with Yap1 to enhance transcription (Sawada et al. 2005, Sawada et al. 2008). Foxa2 is required for formation of the node and notochord (Weinstein et al. 1994, Ang and Rossant 1994).
