Src phosphorylates Runx1
In developing mouse megakaryocytes and T cells, Src tyrosine kinase phosphorylates Runx1 on seven tyrosine residues, three of which are located in the negative regulatory DNA binding domain (Y254, Y258 and Y260), and four in the inhibitory domain (Y375, Y378, Y379 and Y386). Src-mediated phosphorylation negatively regulates differentiation of hematopoietic progenitors. Tyrosine phosphorylated Runx1 exhibits decreased affinity for the Gata1 transcription factor and increased affinity for the SWI/SNF complex (Huang et al. 2012).
