The mammalian canonical BRG1-associated factors (BAF) complex is the paralog of the budding yeast SWI/SNF complex. cBAF is smaller and more abundant than the related polybromo-associated BAF (pBAF) complex. Shared between the two complexes are proteins (or members of protein families) that make up the core of both complexes, including SMARCC and SMARCD proteins, the catalytic subunits SMARCA4 (BRG1) or SMARCA2 (BRM), ACTB, ACTL6A/B and BCL7A/B/C (Wang, Cote et al, 1996; Wang, Xue et al, 1996; Mashtalir et al, 2018; Mashtalir et al, 2020; He et al, 2020; reviewed in Mittal and Robert2, 2020; Cenik and Shilatifard, 2021; Wang and Tang, 2023). Distinguishing components of the cBAF complex include ARID1A or ARID1B, and DPF1/2/3, and SS18 or SS18L1 (shared with ncBAF, described below) (reviewed in Mittal and Roberts, 2020; Cenik and Shilatifard, 2021; Innis and Cabot; 2020; Wang and Tang, 2023).
Mammalian cBAF complexes are required for chromatin remodelling and oppose polycomb repression (Kadoch et al, 2017; Stanton et al, 2017; Wilson et al, 2010). Unlike in yeast, however, mammalian cBAF complexes are able to support both gene activation and repression (Boyer et al, 2005; Ho et al, 2009).
