MITF-M binds to promoter and intronic elements of the E-cadherin (CDH1) to upregulate its expression as assessed by electrophoretic mobility shift and reporter gene assay, ChIP-seq and CUT-and-RUN DNA binding assay (Mansky et al, 2002; Dilshat et al, 2021; Laurette et al, 2015). Consistent with this, protein and mRNA levels of CDH1 are upregulated in melanoma cell lines upon CRISPR-mediated knockdown of MITF-M, as determined by Western and RT-qPCR (Dilshat et al, 2021).
