MITF-M binds to the PXDN gene to repress its expression in melanoma cells. CRISPR- or siRNA-mediated MITF-M knockdown increases expression of PXDN, consistent with a repressive role for MITF-M at this locus. CUT-and-RUN and binding site motif analayses suggest that MITF-M binds directly to the PXDN gene region to repress its expression (Dilshat et al, 2021).
PXDN plays a role in extracellular matrix organization (ECM) and stability through its collagen crosslinking activity (Peterfi et al, 2009; Lee et al, 2020). By regulating the expression of PXDN and other ECM and focal adhesion-related genes, MITF-M impacts the balance of proliferative (higher MITF-M levels) versus quiescent or invasive properties of melanoma cells (lower MITF-M levels), as described by the "rheostat model" (Carreira et al, 2006; Dilshat et al, 2021; reviewed in Goding and Arnheiter, 2019; Rambow et al, 2019).
