Phosphorylated BMAL1:CLOCK heterodimers bind E-box DNA sequences in the isoform-specific promoters of the RORA (RORalpha) gene and enhance transcription (inferred from mouse homologs). RORA, in turn, activates expression of BMAL1 and CLOCK, forming the secondary loop of the circadian clock (reviewed in Cox and Takahashi 2019). RORA also regulates hundreds of other genes, including genes involved in lipid and steroid metabolism (reviewed in Lee et al. 2021).
