Cyclin-dependent kinase 5 (CDK5) phosphorylates PER2 on serine-396 (corresponding to serine-394 of mouse Per2) in the cytosol during the day (inferred from mouse homologs in Brenna et al. 2019). Phosphorylated PER2 interacts with CRY1 (inferred from mouse homologs) and CSNK1D or CSNK1E to form the PER:CRY:kinase complex which, after phosphorylation of PER and CRY proteins, enters the nucleus to negatively regulate the transactivation activity of BMAL1:CLOCK (reviewed in Takahashi et al. 2016).
