Subunits of the base of the 19S regulatory particle (19S RP) of the 26S proteasome, PSMC3 and PSMC6, which are orthologs of yeast Rpt5 and Rpt4, respectively, form a dimer (reviewed in Tomko and Hochstrasser 2013). Binding of endogenous human PSMC3 (Rpt5) and PSMC6 (Rpt4) was demonstrated in the human embryonic kidney cell line HEK293T, and confirmed by using recombinant human proteins (Kaneko et al. 2009). Knockdown or PSMC3 reduces the protein level of PSMC6 and vice versa (Kaneko et al. 2009). Interactions of human PSMC3 and PSMC6 with each other and the surrounding proteins in different 19S RP conformations were structurally characterized by cryogenic electron microscopy (cryo-EM) (Lu et al. 2017).
