Expression of the PLIN4 gene, encoding Perilipin-4, also known as adipocyte protein S3-12, is positively regulated in fat tissue by the PPARG:RXRA heterodimer and KMT2C (MLL3)/KMT2D (MLL4) histone methyltransferase complexes. In human adipocyte cell line SGBS, the expression level of PLIN4 gene correlates with PPARG gene expression (Dalen et al. 2004). In mouse white preadipocyte line 3T3-L1, treatment with PPARG agonists induces Plin4 gene expression (Dalen et al. 2004). PPAR elements (PPAREs) are conserved between human and mouse PLIN4 gene loci, and mouse Plin4 gene reporter construct is activated by exogenously expressed ligand-activated PPARG:RXRA heterodimer (Dalen et al. 2004). Expression of the Plin4 gene is severely reduced in differentiating mouse brown preadipocytes expressing histone H3.3 K4M mutant or Kmt2c (Mll3) and Kmt2d (Mll4) with deleted SET domains (Jang et al. 2019: RNA-seq, supplementary information).
