MITF-M binds to the promoter of the RAB27A gene as determined by EMSA and ChIP in human and mouse cells (Chiaverini et al, 2008, Strub et al, 2011; reviewed in Cheli et al, 2010; Goding and Arnheiter, 2019). MITF-M binding to two E box elements at position-58 to -53 and -47 to -42 drives alpha-MSH and forskolin-responsive expression of RAB27A in mouse and human cells (Chiaverini et al, 2008).
Active GTP:bound RAB27A forms a tripartite complex with MYO5A (another MITF-M target gene) and MLPH (also known as melanophilin) to regulate the localization of melanosomes (Bahadoran et al, 2001; Fukuda et al, 2002; Nagashima et al, 2003; Kuroda and Fukuda, 2004; Alves et al, 2013; Evans et al, 2014; Alves et al, 2017; reviewed in Seabra and Coudrier, 2004; Cheli et al, 2010; Hume and Seabra, 2011). MYO5A interacts with the actin cytoskeleton to localize melanosomes to the ends of dendrites in a manner that also depends on RAB27A and MLPH. Experiments in which MITF-M levels are deliberately manipulated in vitro indicate that MITF-M, by regulating RAB27A, plays a role in melanosome localization (Kuroda and Fukuda, 2004; Chiaverini et al, 2008).
