MITF-M activates the expression of PPARGC1A by binding to E-box elements in its promoter (Haq et al, 2013; Vazquez et al 2013). PPARGC1A is a transcriptional co-activator that has roles in mitochondrial biogenesis and expression of genes involved in oxidative phosphorylation (Mootha et al, 2003; reviewed in Goding and Arnheiter, 2019). Upregulation of PPARGC1A consequently promotes a metabolic shift towards oxidative phosphorylation (Haq et al, 2013; Vazquez et al, 2013; reviewed in Falletta et al, 2022). PPARGC1A has also been identified as a transcriptional activator of MITF-M in concert with SOX10, establishing a positive feedback loop (Shoag et al, 2013).
