The chromodomain of the MPHOSPH8 (MPP8) subunit of the HUSH complex binds trimethylated lysine-9 of histone H3 (H3K9me3) (Kokura et al. 2010, Li et al. 2011, Tchasovnikarova et al. 2015) and recruits the HUSH complex to existing repressive chromatin (Tchasovnikarova et al. 2015). TASOR serves as the central unit of the HUSH complex by binding both MPHOSPH8 and PPHLN1 (Douse et al. 2020). Periphilin (PPHLN1) binds as a dimer to TASOR (Prigozhin et al. 2020). The HUSH complex binds young L1 elements L1HS, L1PA2, L1PA3 (Liu et al. 2018, Robbez-Masson et al. 2018).
