The interaction between the C-terminal fragment of DELE1 and EIF2AK1 (HRI) enhances the phosphorylation of EIF2S1 by EIF2AK1 (Fessler et al. 2020, Guo et al. 2020, Cheng et al. 2022) apparently by increasing the autophosphorylation of EIF2AK1 (Fessler et al. 2020, Cheng et al. 2022). EIF2S1 is a subunit of the EIF2alpha complex, which participates in translation initiation. Phosphorylation of EIF2S1 inhibits general translation but increases translation of specific mRNAs such as DDIT3 (CHOP) and ATF4, that have upstream open reading frames (Fessler et al. 2020, Guo et al. 2020, Cheng et al. 2022).
