5' capping of respiratory syncytial virus (RSV) mRNAs is catalyzed by the polyribonucleotidyl transferase and methyltransferase domains of the catalytic subunit L of the RNA-dependent RNA polymerase (RdRP) complex (Liuzzi et al. 2005). The so-called cap0, labeled m7GpppG- as all RSV transcripts start with guanidine (Liuzzi et al. 2005), is created first, followed by cap methylation and creation of cap1, labeled m7G(5')pppGm- (Liuzzi et al. 2005, Sutto-Ortiz et al. 2021). Cap methylation is not essential for transcription of RSV mRNAs (Liuzzi et al. 2005). The methyltransferase domain of the L protein is required for 2'-O-methylation and creation of cap1, and the complex of L and P proteins is sufficient for the 2'-O-methylation to occur (Sutto-Ortiz et al. 2021).
