The pluripotency factor POU5F1 (OCT4) (Nichols et al. 1998) is expressed in human primordial germ cells differentiated in vitro (Irie et al. 2015, Sasaki et al. 2015, Chen et al. 2018, Fang et al. 2020, Tang et al. 2022). During differentiation of primordial germ cells, SOX17 and TFAP2C bind enhancers of the POU5F1 gene and activate its transcription (Tang et al. 2022, Chen et al. 2018). Absence of SOX17 leads to total loss of POU5F1 expression in embryoids (Irie et al. 2015). PRDM1 may (Irie et al. 2015) or may not (Sasaki et al. 2015) enhance expression of POU5F1. POU5F1 acts with PRDM1 and PAX5 to activate expression of germline genes and repress gene expression in somatic cell lineages (Fang et al. 2020).
