TBX3 binds to the promoter of the CDH1 gene to repress its transcription as assessed by electrophoretic mobility shift assay (EMSA), chromatin immunoprecipitation, reporter gene assays and RT-PCR (Rodriguez et al, 2008). AKT3-dependent phosphorylation of TBX3 promotes its nuclear localization, enhances its repression activity towards CDH1 and may increase its DNA binding affinity (Peres et al, 2015; Peres et al, 2021). TBX3 is overexpressed in melanoma and contributes to its migration and invasiveness, in part through regulation of CDH1 expression (Boyd et al, 2013; Peres and Prince 2013; Peres et al, 2010; Peres et al, 2017; reviewed in Khan et al, 2020).
