The TYRP1 gene encodes 5,6-dihydroxyindole-2-carboxylic acid oxidase. TYRP1 has cAMP-responsive M- and E- box elements in its promoter that are bound by MIITF-M as demonstrated by ChIP and EMSA in mouse and human cells. Transcriptional activity at the TYRP1 promoter is enhanced by the MITF-M-dependent recruitment of a SWI/SNF complex containing SMARCA4 (BRG1), SMARCE1 (BAF57) and SMARCB1 (INI1) (Bertolotto et al, 1996; Bertolotto et al, 1998 a,b; De La Serna et al, 2006; reviewed in Goding, 2000; Cheli et al, 2010; Goding and Arheiter, 2019).
The role of TYRP1 in melanin biosynthesis is somewhat ambiguous. Purified mouse TYRP1 has been shown to catalyze the oxidation of the melanin intermediate DHICA (5,6-dihydroxyindole-2-carboxylic acid) to form melanins, however this activity has not been reproduced with the human protein (Jimenez-Cervantes et al, 1994; Kobayashi et al, 1994; Boissy et al, 1998). Other models suggest that TYRP1 may play a role as a chaperone for TYR or as a buffer against melanin toxicity, however definitive evidence for these or other roles remain to be determined (Kobayashi et al, 1998; Johnson and Jackson, 1992; Luo et al, 1994; Rad et al, 2004; reviewed in Le et al, 2021)
