DCT (also known as TYRP2) encodes L-dopachrome tautomerase, an enzyme involved in melanin biosynthesis that converts L-dopachrome into 5,6-dihydroxyindole-2-carboxylic acid (Jackson et al, 1992; Tsukamoto et al, 1992; Pennamen et al, 2021; Bouchard et al, 1994; reviewed in Cheli et al, 2010). Expression of DCT during melanogenesis is initially repressed through the combined actions of PAX3, LEF1 and TLE4 (Yasumoto et al, 2002; Ludwig et al, 2002; Lang et al, 2005). This repressive complex is replaced by MITF-M and SOX10 binding to cognate elements in the promoter concurrent with the recruitment of CTNNB1 in place of TLE4 (Yasumoto et al, 2002; Ludwig et al, 2002; Saito et al, 2002; Saito et al, 2003; Lang et al, 2005; reviewed in Cheli et al, 2010).
