During gastrulation in mice, Goosecoid (Gsc) is expressed at the anterior end of the primitive streak (Blum et al. 1992) then in the foregut endoderm (Belo et al. 1998). MIXL1 binds elements in the upstream promoter of the GSC gene and activates transcription (inferred from mouse homologs). High NODAL signaling in the anterior primitive streak activates expression of GSC (Xu et al. 2011) through the phosphorylation of SMAD2 and SMAD3, which bind together with SMAD4 at the GSC promoter and activate transcription (inferred from human pluripotent stem cells in Brown et al. 2011, Teo et al. 2011, Danehvar et al. 2016). BMP signaling reduces the expression of GSC (Xu et al. 2011).
