SET1A complex, associated with CXXC1 (CFP1) and WDR82, binds to BOD1L1 (also known as BOD1L) (van Nuland et al. 2013). The N-terminal part of BOD1L1 contains a region with sequence homology to the yeast Shg1, a component of the yeast COMPASS complex, and this region of BOD1L1 is necessary and sufficient for the interaction with SETD1A (Higgs et al. 2018). The SET1A complex bound to BOD1L1 is recruited to stalled replication forks, likely through interaction of BOD1L1 with FANCD2, where SETD1A-catalyzed H3K4 methylation prevents deleterious fork resection (Higgs et al. 2018). In addition, H3K4 methylation by the SET1A complex, recruited to DNA double strand breaks (DSBRs) by BOD1L1, facilitates recruitment of RIF1, thus promoting non-homologous end joining (NHEJ)-mediated repair of DSBRs (Bayley et al. 2022).
