MEN1 (also known as Menin), a tumor suppressor protein mutated in multiple type I endocrine neoplasia, binds to the MLL2 complex. The endogenous complex of MEN1 and MLL2 was co-immunoprecipitated from human cervical carcinoma cell line HeLa and human embryonic kidney cell line HEK293 (Hughes et al. 2004; Jiang et al. 2013: recombinant human KMT2B was used for co-immunoprecipitation from HEK293 cells). The KMT2B (also known as MLL2), similar to the closely related KMT2A (also known as MLL1 or MLL), contains the menin-binding motif (MBM) at its N-terminus, spanning amino acid residues 16–35 (Huang et al. 2012). Based on quantitative studies in HeLa cells, MEN1 has 4- to 5-fold-lower abundance in MLL1/MLL2 complexes than KMT2A/KMT2B, indicating that not all MLL1/2 complexes contain menin (van Nuland et al. 2013).

Huang et al. 2012: recombinant human MEN1 and KMT2B were used.
