The first phase of active DNA demethylation in the male pronucleus is initiated by deamination of cytidine residues by AICDA (AID), which produces T-G mismatches that are repaired by the long patch base excision repair pathway to yield a C-G base pair (inferred from mouse homologs). If the repair patch encompasses a 5-methylcytidine residue adjacent to the T-G mismatch then the 5-methylcytidine is replaced by cytidine, resulting in a loss of DNA methylation (inferred from mouse cells).
