DUX4 binds thousands of LTRs of human endogenous retrovirus-like (HERV-like) retrotransposons, notably the leucine-tRNA primed HERVL family, and MLT2A solo LTRs (Young et al. 2013, De Iaco et al. 2017, Hendrickson et al. 2017, Vuoristo et al. 2022). DUX4 activates bidirectional transcription of the retroelements (Vuoristo et al. 2022), which peaks in cleavage stage embryos (Hendrickson et al. 2017). Many of the LTR elements serve as alternative promoters for genes activated during zygotic genome activation (Young et al. 2013).
