DUX4 binds four sites in the upstream promoter and enhancer region of ZSCAN4 (Geng et al. 2012, De Iaco et al. 2017) and a site in the second exon of the ZSCAN4 gene (Choi et al. 2016) and recruits EP300 (p300) and CREBBP (CBP) (Choi et al. 2016, Vuoristo et al. 2022). DUX4 activates bidirectional transcription of an ERVL-MaLR retrotransposon element 20 kb upstream of the ZSCAN4 transcription start site that upregulates expression of ZSCAN4 (Vuoristo et al. 2022). TPRXL, TPRX1, and TPRX2 are translated at the time of zygotic genome activation and TPRX1 and TPRX2 bind and activate the ZSCAN4 gene (Zou et al. 2022). In mouse, Obox genes appear to be orthologous to human TPRX2 (Maeso et al. 2016) and bind and activate mouse Zscan4 genes (Ji et al. 2023).
