The nucleotidyl transferase in the N-terminal (NiRAN) domain of the SARS-CoV-2 nonstructural protein (nsp12) transfers guanosine-5'-monophosphate onto lysine-2 of nsp7. Nsp7 Lys-2 is well-conserved within the Coronavirinae subfamily. The SARS-CoV-2 RNA-dependent RNA Polymerase (RdRP) structure shows nsp7 Lys-2 located at the interface between nsp8 and nsp12. Similar modiication of nsp8 was observed, with the exact site modified being unknown. Nsp12 possibly also acts as nucleotidyl transferase to mediate viral RNA capping (Conti et al, 2021).
