Upon neuronal activity-induced influx of calcium ions, Ca2+ binds to KCNIP3 homotetramers. Calcium-bound KCNIP3 cannot associate with target DNA sequences (Carrion et al. 1999). Based on studies with mouse Kcnip3, which has amino acid sequence 91% identical to human KCNIP3, Ca2+ binds to two high-affinity sites in Kcnip3 under physiological conditions, when magnesium is present. In the absence of magnesium, Ca2+ can bind to three sites in KCNIP3 (Osawa et al. 2005).
