Based on studies in mice, in the absence of neuronal activity-induced calcium influx, KCNIP3 (DREAM) binds to the promoter of the NPAS4 gene and represses NPAS4 transcription (Mellström et al. 2014). KCNIP3 binds to target promoters as a homotetramer (Carrion et al. 1999). Magnesium is required for stable binding of KCNIP3 homotetramers to target DNA. KCNIP3 possesses one high-affinity, constitutive, Mg2+ binding site and two low-affinity Mg2+ sites (Osawa et al. 2005).
