While phosphorylation of SHC1 upon binding to homodimers of overexpressed ERBB2 has not been tested, it is assumed, based on SHC1 binding and downstream activation of RAS signaling (Pickl and Ries 2009), that ERBB2 homodimers, similar to ERBB2 heterodimers, phosphorylate SHC1, enabling it to recruit GRB2:SOS1 complex and activate downstream RAS/RAF/MAPK signaling.
