MBTPS2 (S2P) cleaves the transmembrane domain of ATF6B near the cytoplasmic face, releasing the N-terminal cytoplasmic domain into the cytosol (Haze et al. 2001, Guan et al. 2009, Odisho et al. 2015). By inference from SREBFs (SREBPs) and ATF6, the cleavage is believed to occur around amino acid residue 399. Glycosylation of 5 sites in the lumenal domain of ATF6B is required for cleavage (Guan et al. 2009).
