USP44 specifically mediates the deubiquitination of CDC20, which promotes the association of MAD2L1 and CDC20, which in turn increases the stabilty of the MAD2L1-CDC20-APC/C ternary complex (Mitotic Checkpoint Complex), thereby preventing premature activation of the Anaphase Promoting Complex/Cyclosome (APC/C) (Stegmeier et al. 2007).
