Mitochondrial brown fat uncoupling protein 1 (UCP1, SLC25A7) transports a proton from the mitochondrial intermembrane space to the mitochondrial matrix, generating heat. The reaction does not occur without activation by fatty acids binding to the transporter (Zhao et al., 2017; reviewed by Li & Fromme, 2022). Phosphatidylethanolamines (PE), but not cardiolipins, in the membrane, are activating (Johnson et al., 2023), and purine nucleotides act as inhibitors (Kang & Chen, 2023). UCP1 can act as a monomer, but homodimer and -tetramer forms exist - we are showing the dimer here. UCP1 is mainly expressed in brown fat (Hoang et al., 2013; reviewed in Bertholet & Kirichok, 2022).
