Homologous recombination in vertebrates is catalyzed by Rad51. Like bacterial RecA, Rad51 polymerizes on single stranded DNA to form nucleoprotein filaments that catalyze pairing and strand exchange with homologous DNA. Two breast cancer susceptible genes seem to regulate this event either directly or indirectly. BRCA1 contributes to DNA repair by regulating recombinase Rad51 assembly in a manner functionally analogous to various Rad51 assembly factors and Rad51 mediators. BRCA1 is also involved in correct localization of BRCA2 to chromatin at damage foci. CHK2 mediated BRCA1 phosphorylation is also required for Rad51 assembly. BRCA2 is a Rad51 mediator that acts to displace RPA and load Rad51 onto ssDNA.
