Upon DNA damage, the inactive ATM dimer auto-phosphorylates, causing dimer dissociation and kinase activation. Activated ATM subsequently phosphorylates Abelson family kinases c-Abl and Arg, which can then phosphorylate Tyr46 of Rad51, leading to the formation of Rad51 nuclear foci.
