The vertebrate Rad17 protein is structurally related to largest subunit of replication factor C (RFC1) and associates with four smaller RFC subunits (RFC2-5). The Rad17-RFC complex, an alternate clamp loader, recognizes the primer-template junction during DNA replication. In a process similar to that occurring in yeast homologs, the Rad17-RFC complex then recruits and loads 9-1-1 complexes to DNA lesions, stimulating subsequent activation of the Chk1 kinase in an early phase of the intra-S-phase checkpoint.
