During early embryonic patterning, Toll (TL) is required for establishing the dorsoventral axis. The TL signal is initiated by activation of its ligand Spatzle (SPZ) by a serine protease cascade. Activation of this cascade is regulated by the probable chaperone protein Windbeutel (WBL) and by Pipe (PIP), a heparan-sulphanotransferase that has been proposed to create a heparin-modified proteoglycan that is required for local activation of the cascade. PIP is the only dorsoventral factor to be expressed on the ventral side of the embryo and is thought to be the key in activating the cascade. The full-length serine protease zymogen, Gastrulation Defective (GD) is cleaved between Arg136 and Lys137. This separates the inhibitory prodomain from the now active mature enzyme. How GD is activated remains uncertain, although a direct interaction with Nudel (NDL) or autoproteolytic processing cannot be ruled out.
