Ptcra gene expression is positively stimulated by Notch3 and inhibited by Ik1

Mouse Notch3 intracellular domain, NICD3, induces transcription from the Ptcra gene promoter, encoding pre-TCR-alpha, in a dose-dependent manner. The Ptcra promoter contains several Rbpj-binding sites and NICD3-mediated induction of Ptcra is Rbpj and Maml dependent (Talora et al. 2003, Bellavia et al. 2007). Binding of Ik1, a splicing isoform of Ikaros (Ikzf1) to the Ptcra promoter competes with Rbpj binding and results in repression of Ptcra transcription (Bellavia et al. 2007).
